Despite the advances of modern medicine, the wound healing by the secondary intention (without regional epithelium contraction) takes considerable time and remains a challenge.

It is necessary to neutralize the powerful oxidative stress, apoptosis, and at the same time enhance tissue regeneration to accelerate the wound healing processes.

The experimental study was conducted to assess the effectiveness of the impact of drugs on the basis of peroxiredoxin 6 (Prx 6) and paracrine factors, mesenchymal stem cells (MSCs) on the healing of an open wound by secondary intention.

Peroxiredoxin 6 is the enzyme, which have strong antioxidant action. Prx 6 is involved in cells protection from oxidative stress. As shown recently, paracrine factors have a strong apoptotic, anti-inflammatory and regeneration properties.

Investigations were carried out on the rates. Simulation of the skin wound healing process provided for excision of skin flap to the fascia in free wool interscapular region of animal. In wounds was used antibiotic gentamicin to prevent bacterial infection.

The experimental animals were divided into four groups depending on the test wound drug: saline; Prx 6 (1 mg / ml); cellulosic hydrophilic gel with MSCs paracrine factors; gel with paracrine factors + Prx 6. Further, in all four groups, collagen film was used on the top of wound surface.

The effectiveness of wounds treatment was performed by histological method. The severity of inflammation, the degree of maturity of the formed granulation tissue, the degree of epithelialization of the wound surface was evaluated.

On the 3rd, 7th and 14th day animals was decapitated for the morphological studies. Terms of removing animals from the experiment corresponded to the phases of wound healing process.

On the first day it is suppressed oxidative stress by Prx 6, which had a crucial influence on the further recovery of the wound defect. The using of gel with paracrine factors of MSCs has a significant positive impact, reinforcing the regenerative processes.

The using of combination of two drugs shows the best picture of the recovery. Applying the gel with paracrine factors of MSCs with Prx 6 can significantly increase the rate of wound healing. Thus, antioxidant effect of peroxiredoxin 6 and regenerative effect of MSCs growth factors was shown on the damaged skin tissue.
